Aldolase adalah enzim yang bekerja tanpa bantuan oksigen dan terlibat dalam reaksi glikolisis yaitu reaksi pemecahan glukosa menghasilkan energi. Aldolase bekerja mempercepat(katalis) reaksi kimia organik bolak-balik. Pada salah satu tahap reaksi glikolisis, enzim ini mengubah satu molekul fruktosa 1,6- difosfat (HDP) menjadi dua molekul yang berbeda yaitu, satu molekul gliseraldehid-3-fosfat (G3P) dan satu molekul dihidroksiaseton fosfat (DHAP). Keduanya memiliki struktur yang berbeda, tetapi pada akhirnya dihidroksiaseton fosfat akan diubah menjadi gliseraldehid-3-fosfat.




Jenis-jenis


Ada 3 macam aldolase yaitu aldolase A, aldolase B, dan aldolase C. Ketiga jenis enzim aldolase ini merupakan produk dari bagian gen yang berbeda. Aldolase A mengkatalis senyawa yang terlibat dalam reaksi glikolisis, yaitu fruktosa 1,6 bifosfat, banyak terdapat pada jaringan otot. Aldolase B berkerja pada glukoneogenesis, banyak terdapat pada hati. Sedangkan aldolase C merupakan senyawa peralihan antara aldolase A dan aldolase B, banyak ditemukan pada enzim otak yang spesifik di sel Purkinje. Bila diteliti secara elektroforesis ketiga enzim ini sebenarnya serupa.




Cara kerja


Aldolase bekerja dengan dibantu oleh enzim dehidrogenase. Kerjasama enzim aldolase dengan dehidrogenase mengkatalis dua tahap terakhir pemutusan katekol, toluene, naftalena, dan senyawa aromatik lainnya. Keduanya juga mendegradasi 4-hidroksi-2-ketovalerat menjadi piruvat dan asetil KoA, dengan asetaldehid sebagai senyawa intermedietnya. Kedua enzim ini berasosiasi sangat erat satu sama lain, di mana tanpa dehidrogenase, aldolase akan menjadi inaktif dan dehidrogenase memiliki aktivitas khusus bila tidak ada enzim aldolase. Reaktivitas enzim aldolase bersifat termodulasi secara alosterik dengan mengikat dehidrogenase sebagai kofaktor pada sisi aktifnya, jadi aktivitasnya dirangsang dengan penambahan dehidrogenase.




= Fermentasi Asam Laktat

Terdapat dua jenis bakteri fermentasi asam laktat, yaitu bakteri homofermentatif dan bakteri heterofermentatif. Perbedaan keduanya terdapat pada ada atau tidaknya enzim aldolase. Bakteri homofermentatif memiliki enzim aldolase sehingga dapat memproduksi dua laktat dari glukosa yaitu 2-Gliseraldehid-3-fosfat dan Dihidroksiaseton fosfat. Sementara itu, bakteri heterofermentatif tidak memiliki enzim aldolase sehingga tidak dapat memecah fruktosa bifosfat menjadi triosa fosfat. Namun, bakteri ini dapat mengoksidasi glukosa-6-fosfat menjadi 6-fosfoglukonat yang kemudian didekarboksilasi menjadi pentosa fosfat.
