Pulsed Field Gel Electrophoresis (PFGE) adalah metode yang digunakan untuk memisahkan DNA yang berukuran besar. Metode ini pertama kali diperkenalkan pada tahun 1984 oleh Schwartz dan Cantor. Metode PFGE ini berbeda dengan elektroforesis agarosa konvensional karena orientasi dari medan elektrik memiliki periode dengan teratur. Keberagaman medan listrik inilah yang menyebabkan DNA yang berukuran besar dapat terpisah.




Prinsip


Prinsip dari PFGE ini adalah dengan adanya pergantian voltase di setiap elektrode tersebut, maka DNA yang berukuran besar dapat terpisah dimana fragmen DNA yang berukuran kecil akan berjalan terlebih dahulu karena memiliki waktu orientasi yang lebih sedikit dibandingkan dengan fragmen DNA yang berukuran besar yang membutuhkan waktu orientasi yang lebih lama.




Aplikasi


Teknik PFGE ini dapat digunakan untuk menganalisis kekerabatan di antara galur bakteri yang berbeda namun masih dalam satu spesies dan menjadi metode yang sangat efektif dalam mengestimasi ukuran genom serta mengkonstruksi peta restriksi dari suatu spesies.
